چکیده

Pleiotropy, a situation in which a single gene influences multiple phenotypic traits, can arise in a variety of ways. This paper discusses possible underlying mechanisms and proposes a classification of the various phenomena involved.

Pleiotropic costs of niche expansion in the RNA bacteriophage phi 6.

Natural and experimental systems have failed to universally demonstrate a trade-off between generalism and specialism. When a trade-off does occur it is difficult to attribute its cause to antagonistic pleiotropy without dissecting the genetic basis of adaptation, and few previous experiments provide these genetic data. Here we investigate the evolution of expanded host range (generalism) in th...

On the definition and measurement of pleiotropy.

Pleiotropy, a 103-year-old concept [1], refers to the phenomenon of one mutation affecting multiple traits. Compared with the fundamental importance of pleiotropy in genetics, development, evolution, and medicine, our empirical knowledge of the prevalence and mechanism of pleiotropy is scarce [2]. Given this context, Paaby and Rockman’s recent Opinion article on pleiotropy and its empirical est...

Continuous inflation analysis: a threshold-free method to estimate genetic overlap and boost power in imaging genetics

Methods to quantify genetic overlap may elucidate relationships between disparate traits, and provide Bayesian priors to guide the search for genetic influences on brain measures. Here we describe a threshold-free method called continuous inflation analysis (CIA), which we used to compare genomewide association statistics (GWAS) for the volumes of eight brain regions, computed from brain MRI. O...

Genepleio Software for Effective Estimation of Gene Pleiotropy from Protein Sequences

Though pleiotropy, which refers to the phenomenon of a gene affecting multiple traits, has long played a central role in genetics, development, and evolution, estimation of the number of pleiotropy components remains a hard mission to accomplish. In this paper, we report a newly developed software package, Genepleio, to estimate the effective gene pleiotropy from phylogenetic analysis of protei...

Toward a molecular understanding of pleiotropy.

Pleiotropy refers to the observation of a single gene influencing multiple phenotypic traits. Although pleiotropy is a common phenomenon with broad implications, its molecular basis is unclear. Using functional genomic data of the yeast Saccharomyces cerevisiae, here we show that, compared with genes of low pleiotropy, highly pleiotropic genes participate in more biological processes through di...
